Objetivos de aprendizagem (conhecimentos, aptidões e competências)
A Biologia Molecular estuda os mecanismos celulares relacionados com a replicação do material genético e com a síntese de proteínas, incidindo nas interações entre o ADN, ARN e proteínas na regulação da expressão genética; a estrutura e topologia dos ácidos nucleicos, os mecanismos de replicação, transcrição e tradução do material genético e sua regulação, o código genético, as mutações e os mecanismos de reparação e recombinação do ADN. Bem cmo, conhecimentos sobre bioinformática de modo a ser capaz de utilizar as bases de dados mais relevantes para a área da Biologia Molecular. Nas aulas práticas serão executados trabalhos que permitirão aos alunos desenvolver a sua capacidade prática no que se refere aos métodos de isolamento e análise dos ácidos nucleicos. Deverão ainda consolidar as suas capacidades de proceder ao registo adequado dos resultados obtidos e a elaboração de relatórios científicos, demonstrando a capacidade de análise e interpretação dos resultados obtidos.
Conteúdos programáticos
Introdução à Biologia Molecular. 2. Estrutura dos ácidos nucleicos; genoma procariota e eucariota; estrutura da cromatina; estabilidade dos ácidos nucleicos; genoma mitocondrial. 3. Expressão genética: Transcrição; RNA polimerase; estrutura de genes procariotas e eucariotas; processamento pós-transcrição. 4. Transporte nuclear. 5. Tradução (de RNA a proteína): O código genético; tradução em procariotas e eucariotas; modificações pós-tradução; destruição programada de proteínas. 6. Regulação da expressão genética: Proteínas de ligação ao DNA;. 7. Replicação, reparação e recombinação de DNA. 8. Manipulação de DNA e de genes. 9. Tecnologias básicas; sequenciação; PCR, clonagem; aplicações de tecnologia de DNA recombinante; bioinformática. 10. RNA de interferência.
Metodologias de ensino (avaliação incluída)
Os principais conceitos teóricos são apresentados ao estudante através da metodologia expositiva, com recurso ao suporte audiovisual. A metodologia interrogativa é utilizada ao estimular no estudante o interesse e espírito crítico acerca dos casos clínicos apresentados. A metodologia ativa aplica-se na componente prática, em que estudante apresenta o trabalho prático, executa-o de forma mais autónoma possível e interpreta/discute os resultados obtidos.
A avaliação compreende um teste teórico escrito, que corresponde a 60% da classificação final, e um teste prático escrito, que equivale a 40% da nota final. A aprovação da prática está dependente da presença mínima de 80% das aulas. Estão dispensados de exame os alunos com classificação ? 9,5 valores em cada componente.

The students intended learning outcomes (knowledge, skills and competences)
Molecular biology studies the cellular mechanisms related to the replication of the genetic material and protein synthesis, focusing on interactions among DNA, RNA and proteins in the regulation of gene expression. Students should acquire knowledge about the structure and topology of nucleic acids, replication mechanisms, transcription and translation of the genetic material and its regulation, the genetic code, mutations and mechanisms of repair and recombination DNA. They should also acquire basic knowledge of bioinformatics in order to be able to use the most relevant databases to the area of ??molecular biology. In practical classes work will be performed that will enable students to develop a practical capacity as regards the methods of isolation and analysis of nucleic acids. They should also strengthen their ability to make the appropriate record of the results and preparation of scientific reports, demonstrating the capacity of analysis and interpretation of results.
Syllabus
Introduction to Molecular Biology. 2. The genome: structure of nucleic acids; genome prokaryotic and eukaryotic; chromatin structure; Stability of nucleic acids; mitochondrial genome. 3. Gene Expression: Transcription; RNA polymerase; structure of prokaryotic and eukaryotic genes; transcription in prokaryotes and eukaryotes; post-transcriptional processing; 4. Nuclear transport. 5. translation (RNA protein): The genetic code; translation in prokaryotes and eukaryotic; protein synthesis; post-translational modifications; destruction programmed proteins. 6. Regulation of gene expression, DNA binding protein; regulation in eukaryotes. 7. replication, repair and recombination of DNA. 8. DNA and gene manipulation. 9. Basic Technologies; sequencing; PCR, cloning; Recombinant DNA technology applications; the use of bioinformatics. 10. RNA interference.
Teaching methodologies (including evaluation)
In the theoretical classes the material is exposed using audio-visual support and discussion of the presented concepts. The students are encouraged to achieve the pre-established objectives for each concept. This component is evaluated through one written test, with a minimum grade of 9.5 values (CT).
The practical component consists in an initial part of the work-protocol presentation followed by the practical application of the protocol by students, as independently as possible, and the interpretation and discussion of obtained results. The approval to the practical part is dependent on the assistance to a minimum of 80 % of the classes. This component is evaluated through a written test (CP).
The student is excluded from the exam with ? 9.5 values in each component. The final classification (CF) is the average of CT ( ? 9.5 , 60%) and CP ( ? 9.5 , 40%)
