II. DESENVOLVIMENTO EXPERIMENTAL
6. Conclusões
Neste estudo, identificaram-se algumas fluoroquinolonas que in vitro actuam como inibidores potentes e/ou moderados do crescimento e indutoras de citotoxicidade em promastigotas de L.
infantum, reforçando o potencial da investigação de agentes terapêuticos candidatos que
tenham como alvo as topoisomerases destes parasitas. A elevada eficácia antiparasitária observada na balofloxacina, gemifloxacina e clinafloxacina, bem como a eficácia significativa da sitafloxacina, garenoxacina, sparfloxacina e grepafloxacina apoiam a continuação da avaliação das fluorquinolonas e dos seus derivados como alternativas terapêuticas válidas, a ser testadas ao nível molecular, in vitro, in vivo (modelo animal), e em última fase, em testes clínicos.
Verificou-se a exequibilidade da cultura axénica de promastigotas de L. infantum com o recurso a meio de cultura RPMI 1640 suplementado com 10% de soro fetal bovino, 5% de

hemina e 10% de urina humana masculina. Este é um meio de cultura de preparação mais simples e barata que outros meios tradicionalmente usados na cultura de promastigotas. Demonstrou-se ainda a possibilidade de recurso a meios de baixa tecnologia para a incubação das culturas.
O violeta de genciana (trifenilmetano) demonstrou grande acção leishmanicida e pode ter potencialidade como um recurso adicional no tratamento das lesões cutâneas de leishmaniose humana e canina.
