Este trabalho demonstrou que a atividade da enzima NTPDase pode ser modificada em plaquetas de pacientes hipertensos. No entanto, mais estudos são necessários para melhor entender os mecanismos envolvidos na possível diminuição da hidrólise dos nucleotídeos ATP e ADP e conseqüente alteração no processo trombótico. Esperamos que esses resultados representem uma importante contribuição para estabelecer a associação dos fármacos com os processos fisiológicos.

TERMO DE CONCENTIMENTO LIVRE E ESCLARECIDO 1. Título

Interferência de vasodilatadores na hidrólise dos nucleotídeos da adenina em plaquetas de pacientes hipertensos.

2. Objetivos

a. Verificar a interferência dos vasodilatadores arginina, nitroprussiato de sódio, hidralazina e captopril, na atividade da enzima NTPDase em pacientes hipertensos e saudáveis;

b.Determinar a cinética da enzima NTPDase frente ao vasodilatador que apresentar maior influência inibitória;

3. Registro

O estudo será desenvolvido no Centro de Ciências Naturais e Exatas, Departamento de Química, Programa de Pós-Graduação em Bioquímica Toxicológica, no Laboratório de Enzimologia Toxicológica, da Universidade Federal de Santa Maria. O presente estudo envolverá pacientes hipertensos vinculados ao Programa de Assistência aos Pacientes Diabéticos, Hipertensos e Diabéticos- Hipertensos da Secretaria Municipal de Saúde do município de Cruz Alta, RS. Esse estudo com voluntários humanos obteve a aprovação junto a Comissão de Ética do Centro de Ciências da Saúde da Universidade Federal de Santa Maria, com protocolo n° 013/2004.

4. Procedimento

Os pacientes serão submetidos a uma punção venosa com sistema vacutainer. O material biológico, sangue, será destinado para análise de plaquetas, soro e plasma para determinações bioquímicas e de atividade enzimática. As plaquetas serão analisadas no Laboratório de Enzimologia Toxicológica, Centro de Ciências Naturais e Exatas, Departamento de Química, Programa de Pós- Graduação em Bioquímica Toxicológica, da Universidade Federal de Santa Maria, RS.

5. Riscos individuais

Os pacientes que voluntariamente se submeterem às punções venosas, poderão em casos de coleta com procedimento errôneo desenvolver flebite, flebotrombose, hematoma local, petéquias.
