2.3.3 Sistema de metabolização exógena
As linhagens de S. Typhimuriumsão incapazes de metabolizar compostos químicos via citocromo P450, como acontece nos mamíferos, sendo assim, foi necessária a inclusão de um sistema de metabolização exógena no teste de Ames para permitir a detecção de compostos que precisam ser metabolizados para se tornarem ativos, como as aminas aromáticas, os hidrocarbonetos policíclicos aromáticos e aflatoxinas (AMES et al., 1973b). As enzimas do citocromo P450 geralmente são expressas no fígado, por isso, Malling em 1971, propôs a utilização de um homogenato de fígado de rato para realizar metabolização de compostos químicos que são reativos com o DNA somente após esse processo.
Assim, o sistema de metabolização exógena comumente utilizado é a fração microssomal S9, proveniente de fígado de rato Sprague Dawley previamente tratado com a solução de bifenil policromida (Aroclor 1254) para induzir um aumento de enzimas neste órgão. Para o preparo da fração são acrescidos co-fatores como glicose-6-fosfato e ß-nicotinamida adenina dinucleotídeo fosfato (NADP), além de tampão fosfato, água, cloreto de potássio e cloreto de magnésio (MORON e AMES, 1983). 3. OBJETIVOS 3.1 Objetivo Geral
Avaliar a aplicação do teste de Ames no estudo do potencial mutagênico de plantas medicinais e novas moléculas com atividades biológicas.
3.2 Objetivos específicos
- Relatar a aplicação do teste de Ames na avaliação do potencial mutagênico de plantas medicinais;
- Listar as espécies de plantas utilizadas na medicina popular que já foram avaliadas pelo teste de Ames;
- Investigar o potencial mutagênico de dois novos complexos de cobre(II) com ligantes 2-tiouracil;
- Avaliar a atividade antifúngica dos dois novos complexos de cobre(II).
