1Summary
Ca2+
ions areimportant
second messengers involved in many cellular processes.Signals
are transmittedby raising
thenormally
very low intracellularCa2+
concentration(about
100nM) by
several orders ofmagnitude.
Different molecules, likeCa2+
Channels,antiporters
or ATP driven pumps, areinvolvedin thegeneration
andthe regulationoftheCa2+
signal.
The
plasma
membraneCa2+-ATPase (PMCA)
isinvolved in the restoration of theCa2+ resting
level. Dueto itshigh Ca2+ affinity
itwasproposed
toberesponsible
for the finetuning
of the intracellularCa2+
concentration. The enzyme is encodedby
four genes(PMCA1-4),
which result in four distinct isoforms. Their numberisfurther increasedby
alternativesplicing
at three distinctsites.Analyzing
the tissue distribution of the four human PMCA transcripts and their relative amountsby quantitative PCRon reversetranscribed mRNA, it becomes obvious thatthere exist two differentexpression
patternsfor the four genes. Two geneproducts
were found to beexpressed ubiquitously
in all tissues(PMCA1
and4),
whereasthe othertwo(PMCA2, 3)
weredetected in neuronal andmuscle tissuesonly.
Thehighest
amounts of PMCAmRNAwere found in neuronal tissues. In further
experiments
the variants at the splicesiteswereanalyzed by
RT-PCR,andresultedinthediscovery
ofnewsplice
forms.Aquantitativeanalysis
ofthesplice
formswasperformed
in the different tissues, showing that there exists ageneral,
normally most abundant splice variant at each splice site. Since, no information was available about the different genes and their location at the protein level, antibodiesweregenerated
specificfor the fourgeneproducts.
Thereforethe N-terminalpartsofthefour isoformswereoverexpressed
inE.coli and used to immunize rabbits. The antibodies werehighly specific
for thecorresponding
isoform and showed nocrossreactivity.
All antibodiesrecognized
thecorresponding
rat isoforms, except theantibody against
PMCA4. The antibodies were used toanalyze
the distribution of the four enzymes in different human and rat tissuesby
Western blotanalysis.
Theresultsverifiedthe
finding
obtainedby
theprevious study
on thetranscripts,though
some minordiscrepancies
in theexpression
pattern were found.PMCA1 and 4 were found to be expressed in almost all tissues, whereas PMCA2 and3 werefound again onlyin neuronal tissues. In
general
PMCA1was the more abundant
housekeeping
enzyme with someexceptions
e.g.erythrocytes.
As also shown on thetranscript level, thehighest
amounts ofPMCA were found in neuronal tissues, therefore cellular and subcellular distribution of the PMCA isoforms were
analyzed
in brainby immunohistochemistry
and immunoelectronmicroscopy. Interestingly,
thehousekeeping
isoform(PMCA1)
wasfound tobe expressedubiquitously
in brain but in different relative amounts, whereas PMCA2 and 3 were foundonly
in veryspecific regions (PMCA2: Purkinje
cell dendrites; PMCA3:choroid
plexus).
Thespecific
distribution of theisoforms and their distinct biochemicalproperties
suppose thatthey
represent welladapted
forms forspecific regions containing
different but well definedphysiological
properties.

Kalziumionen sind
wichtige
sekundare Botenstoffe, welche an etlichen zellularen Prozessenbeteiligt
sind. DieSignale
werden durch dieVeranderung
der intrazellularen lonenkonzentration ubermittelt. Die normale Konzentrationliegt
bei zirka 100 nM und kann sich bei derSignalubermittlung
bis auf 800 nM erhohen. Verschiedene Molekiile, wie KalziumionenkanaleOder ATPangetriebene Kalziumionenpumpen
sind fur die ProduktionundRegulation
diesesSignales
verantwortlich.Die Plasmamembran Kalzium-ATPase
(PMCA)
istpartiell
verantwortlich fur dieWiederherstellung
der Grundkonzentration der Kalziumionen. Da dasEnzym
eine sehr hohe Kalziumaffinitat besitzt, wird angenommen, dass es fur dieFeinregulation
der intrazellularen Kalziumionenkonzentrationzustandig
ist. DasEnzym
wird durch vierGenekodiert. Die Variability wird weiter erhohtdurch alternativesSpleissen
an drei Orten.Die
Gewebeverteilung
derTranskripte
der vierGenprodukte
der Plasmamembran Klazium-ATPase und deren relativeMenge
wurde mittels derPolymerase-Kettenreaktion (PCR)
an revers transkribierter mRNA bestimmt. Imgenerellen
wurden zwei unterschiedlicheVerteilungsmuster gefunden:
Die Isoformen 1 und 4(PMCA1, 4)
wurden injedem
Gewebeexprimiert,
wahrend Isoformen 2und 3nurin neuronalemGewebe in hohen Mengen gefundenwurden. Dieswarauchdas Gewebe, in demdie hochsteMenge
an PMCAspezifischer
mRNA dedektiert wurde. In weiterenExperimenten
wurde dasSpleissmuster
an alien drei Orten, an denen alternativesSpleissen
entstehen kann, untersucht. Dabei wurden bisher unbekannteSpleissformen
entdeckt. Diequantitative Analyse
derSpleissformen zeigte,
dasses anjedem
Ort eineHauptform gibt,
welchein jedem Gewebe vorhanden ist und normalerweise denHauptanteil
darstellt.In neuronalem Gewebe wurdeeine sehr hohe Varibialitatvon
Spleissformen gefunden.
Daes keine Informationen uber die Prasenz und die
Verteilung
derProteine der vier PMCA-Formengab,
wurden Isoformenspezifische Antikorper hergestellt.
Dazu wurde der N-terminale Bereich derjeweiligen
Isoform inE.coli
uberexprimiert
und Kaninchen mit demaufgereinigten Peptid
immunisiert. Die
Antikorper
waren hoch isoformen-spezifisch undzeigten
keineKreuzreaktion mit denanderen Isoformen.AlleAntikorper,ausserdem gegen PMCA4, erkannten auch die
entsprechende
Rattenisoform. Die Antikorper wurden inWestern-blot-Experimenten
verwendet urn die vierIsoformen in menschlichen wie auch
Rattengeweben
nachzuweisen. DieResultate entsprachen im Prinzip denErgebnissen,die bei der
