Zusammenfassung
Der
Organismus
Pseudomonas oleovoransverfügt
über einEnzymsystem,
das
befähigt ist,
1-Octen zu7-Octen-l-ol zuhydroxylieren.
Des Weiterenwird die terminale
Doppelbindung
derartoxidiert,
dass1,2-Epoxyoctan
undCaprylaldehyd
entstehen.Während derAldehydbildung
tritt eine intramole¬kulare
1,2-Wasserstoffwanderung
auf. Es istbekannt,
dass dasEpoxid
nichteineZwischenstufefür die
Gewinnung
desAldehyds
darstellt. In der vor¬liegenden
Arbeitwurderegioselektiv
monodeuteriertes 1-Octenmitganzen Pseudomonasoleovorans-Zelleninkubiert,
um den stereochemischen Ver¬lauf der
Bildung
vonCaprylaldehyd festzulegen.
Esgelang nicht,
dener¬wünschten
Caprylaldehyd
in derKulturlösung
nachzuweisen. Eine in den Bakterienzellen anwesendeAldehyddehydrogenase
ist in derLage,
denen-zymatisch gebildeten Aldehyd sogleich
zurCarbonsäurezuoxidieren. Aus diesem Grunde wurde ebenfalls nach derCaprylsäure
Ausschaugehalten.
Aber auchdieses
Metabolisierungsprodukt
konnte nicht erfasst werden. Auf Grund dieses Befundes mussten die relevantenKomponenten
desEnzym¬
systems
derPseudomonas oleovorans isoliert werden.Die
Isolierung
des Rubredoxins und diepartielle Reinigung
der Pseudomo¬nas
oleovorans-Monooxygenase
warenVoraussetzungen,
um einEnzym¬
system zu
erstellen,
das ausNADPH, Spinat-Ferredoxin-NADP+-Reduk-
tase, Rubredoxin
(bzw. Ferredoxin)
und derpartiell gereinigten Monooxy¬
genase bestand. Wurde 1-Octen als Substrat für dieses
POM-Enzymsystem eingesetzt,
konnte dieCaprylsäure
alsOxydationsprodukt
identifiziert wer¬den. Bei
Verwendung
voncis-[l-2H]-l-Octen
als Substrat wardie isolierteCaprylsäure
am ZentrumC(2)
monodeuteriert. Da derDeuterierungsgrad
des Produktes demDeuteriumanteil in
cis-Stellung
desSubstratesentsprach,
warausschliesslichder
cis-ständige Ligand
vonC(l)
nachC(2) gewandert.
Dabei wirdder
primäre Deuteriumisotopeneffekt
eherüberwunden,
alsdassein Wasserstoffatom aus der
trans-Lage
wandern würde. Diese Aus¬schliesslichkeit der
1,2-Wanderung
descis-ständigen Liganden
konnte ineiner
komplementären
Inkubation durch Einsatz vontrans-[l-2H]-l-Octen
als Substrat
belegt

Umden stereochemischen Verlauf der
Aldehydbildung
beschreibenzukön¬nen, musste zudem die
Konfiguration
desWanderungsterminus
untersuchtwerden. Dazu wurde die
Synthese
vonselektiv a-monodeuterierterCapryl¬
säurebekannter
Konfiguration
amC(2)-Zentrum notwendig.
Durch Reduk¬tionvon
[1-2H]-Heptanal
mit dem chiralenMidland-Reagenz
wurdestereo¬selektiv monodeuterierter
[l-2H]-Heptanol hergestellt.
DieKonfiguration
des entstandenen Carbinolzentrums wurde auf
unabhängige
Weise enzyma- tisch undNMR-spektroskopisch
bestimmt. In mehrerenSyntheseschritten
mit bekanntem stereochemischen Verlauf wurde die
[2-2H]-Caprylsäure
hergestellt.
Nach einerVeresterung
der Carbonsäure mitoptisch
reinem L-Mandelsäuremethylester
konnte dieKonfiguration
desC(2)-Zentrums 2H- NMR-spektroskopisch
ermittelt werden. Dieseanalytische
Methodik er¬möglichte
nun, dieKonfiguration
derenzymatisch gebildeten [2-2H]-Ca- prylsäure
zubestimmen.Bei der
Verwendung
von[2-2H]-1-Octen
als Substrat des beschriebenenPOM-Enzymsystems
wurdenachgewiesen,
dass 63% der a-monodeuterier¬ten
Caprylsäure
die(2R)-Konfiguration
besitzen. Die restlichen37% weisen dieantipodale Konfiguration
auf. Wurdecis-[l-2H]-l-Octen enzymatisch
umgesetzt, standen26% der(2R)-konfigurierten
Carbonsäure 74% der(S)- [2-2H]-Caprylsäure gegenüber.
Daaus beidenInkubationsexperimenten
der[2-2H]-Caprylaldehyd entsteht,
wird keiner der Anteile durch die Oxidation zurCarbonsäure verändert. Somit stellt sich dieseVerteilung
bereits bei derBildung
desCaprylaldehyds
ein.Die Ausschliesslichkeit der
1,2-Wanderung
descis-ständigen
Substituentenerlaubt,
dieRichtung
deselektrophilen Angriffs
der oxidierenden Eisen-oxo-spezies
auf dieSubstratdoppelbindung
und dieKonfiguration
desa-monodeuterierten
Caprylaldehyds
miteinander zu korrelieren. Somit ent¬spricht
das(2R)/(2S)-Verhältnis
imAldehyd
derHäufigkeit
deselektrophi¬
len
Angriffes
ausdemjeweiligen
Halbraum derSubstratdoppelbindung.
Alsinterne
Standardisierung
wurdejeweils
dieMenge
derenzymatisch gebilde¬
ten, terminal
ungesättigten Caprylsäure
verwendet. Diese Carbonsäure ist durchHydroxylierung
undOxydation
derSubstratmethylgruppe gebildet
worden.

Da dieses Meta¬bolisierungsprodukt
nicht imgleichen
zellfreienSystem nachgewiesen
wer¬den
konnte,
wurdendie relativen Anteile derkonfigurationsisomeren,
mo-nodeuterierten
Epoxide
aus Inkubationen von eis- bzw.trans-[l-2H]-l-
Octenmit ganzenPseudomonas oleovorans-Zellenermittelt.
Basierendauf denausden Inkubationen mit den
Enzymsystemen
und denganzenBakterienzellengewonnenen Resultaten wurdeein mechanistisches Bild
entworfen,
dasdieBeschreibung
derMetabolisierung
vonterminalun¬gesättigten
Kohlenwasserstoffen mit demEnzymsystem
der Pseudomonas oleovoranserlaubt. StellvertretendwirdderReaktionsmechanismusfür dieMetabolisierung
voncis-[l-2H]-l-Octen
im Schema Adargestellt.

Inanattempttoinvestigate
thestereochemistry
of thealdehyde
for¬mation
regioselectively
monodeuterated 1-octenes were incubated with whole cells of Pseudomonas oleovorans. In this incubationnooctanal could bedetected. Thereforeacellfreeenzymesystem
wasprepared.
Isolation of rubredoxin and
partially purification
of the Pseudomonas oleovorans monooxygenase allowedtoreconstituteanenzyme systemcon-sisting
ofNADPH, spinach-ferredoxin-NADP+-reductase,
rubredoxin(respectively ferredoxin)
and thepartially purified
monooxygenase.Using
1-octeneasSubstratefor this
POM-enzyme
system, octanoic acidwas de- tectableby GLC/MS.
Incubation ofcis-[l-2H]-l-octene yielded
an a-mo-nodeuterated octanoic acid
having
thesamedeuteriumcontentas found incis-position
of the Substrate. Thisimplies
thatonly
theligand
incis-position
of the Substrate is able to
undergo
the1,2-migration
inspite
ofanadverseisotope
effect. Thecomplementary
incubationusing trans-[l-2H]-1-octene
confirmedthe observed
specificity
ofthe1,2-shift.
To
investigate
thestereochemisty
of thealdehyde
formationanalytical
toolshadto be established in order todetermine the
configuration
at theC(2)-
centerof octanalorof octanoic acid. The
synthesis
of[2-2H]-octanoic
acidof known
configuration
was initiatedby
the reduction of[l-2H]-heptanal
witha chiral Midlandreagent. The
configuration
oftheresulting [1-2H]-1- heptanol
wasestablishedby
meansofenzymatic oxidation/reduction
in thepresence ofyeast alcohol
dehydrogenase
andby
NMRanalysis.
Differentconsecutive
synthetic
steps of known stereochemical course were carried out to form the[2-2H]-octanoic
acid of knownC(2)-configuration.

The exclusive
migration
of theligand

of the Substrate double bondallowstocorrelatethe side ofelectrophilic
attackonthe doublebondby
the oxidativeiron-oxo-species
and theconfiguration
of the a-monodeute- ratedaldehyde.
Therefore thequotient (2R)/(2S)
ismonitoring
the ratio ofthe
Re/Re-
andSi/Si-attack
on the Substrate double bond. The amountof 1-octenoic acidproduced by hydroxylation
and oxidation of the Substratemethyl
group allowstodetermine the turnoverof the reaction. Because in¬cubation time is
kept
constant the results from incubation of each of the monodeuterated Substratescanbecompared.
Afurther
product
of theenzymatic
conversionof 1-octene with whole cells of Pseudomonas oleovorans is1,2-epoxyoctane.
Thisepoxide
couldnotbe isolatedusing
thecell free POM enzymesystem.The relative distribution of Constitution isomeric monodeutratedepoxides
was determinedby
incuba¬tion of eis- and
trans-[l-2H]-1-octene
with the bacterial cells.Onthe basis of the results with the POM enzyme system and the
experi-
mentswith whole Pseudomonas oleovorans cells a mechanistic scheme is
proposed
todescribe the metabolism ofterminally
unsaturatedhydrocarbons
