Aspectos biológicos das

leveduras do gênero

Candida

isoladas de candidíase vaginal

Foram estudadas 120 amostras de secreção vaginal coletadas de mulheres com suspeita clínica de candidíase na cidade de Goiânia, GO, identificando-se 57 (47,5%) Candida albicans, 4 (3,3%) Candida krusei e 2 (1,7%) Candida tropicalis. As características morfofisiológicas das leveduras analisadas mostraram células arredondadas e ovais com brotamento unipolar e multipolar e a formação de tubos germinativos em soro fetal bovino. No microcultivo, em ágar

c orn-meal acrescido de tween 80, foram observados pseudomicélios, blastósporos e clamidósporos em cepas de Candida albicans. Em caldo Sabouraud dextrose, presenciou-se a formação de resíduo e sedimento. As colônias g i g a n t e s d e C a n d i d a a p r e s e n t a r a m - s e branco-amareladas, opacas e lobuladas. Topograficamente, notou-se colônias de centro e periferia lisa, rugosa e franjada. A assimilação e fermentação de açúcares ocorreram de acordo com cada espécie de Candida. A produção de proteinase foi detectada em todas amostras de leveduras analisadas e de fosfolipase apenas em Candida albicans. Na análise fenotípica das leveduras obteve-se 9 morfotipos (0000, 3220, 5220, 5230, 5240, 5320, 5330, 5530 e 7330) e 3 biotipos (337, 677 e 777) diferentes. As 56 (98,2%) cepas de Candida albicans foram classificadas como sorotipo A. Todas as espécies de Candida apresentaram-se sensíveis à anfotericina B e nistatina.Cand id a alb ic ans

mostrou-se sensível ao cetoconazol (35,0%), fluconazol (22,8%) e itraconazol (47,4%).

Candida kruseie Candida tropicalisnão foram sensíveis, respectivamente, ao fluconazol e cetoconazol. Através do teste "kendall rank correlation" verificou-se a associação entre a produção de exoenzimas pelas cepas de Candida

e os resultados dos testes de susceptibilidade in vitroà drogas antifúngicas.

A total of 120 samples of vaginal secretion were collected from women with a clinical suspicion of candidiasis treated in the city of Goiânia, GO, Brazil. The study identified 57 (47.5%) strains of Candida albicans, 4 (3.3%) of

Cand id a krusei and 2 (1.7%) of Cand id a tropicalis. Morphophysiological analysis of the yeasts revealed round and oval cells with unipolar and multipolar budding. The formation of germ tubes in bovine fetal serum was also detected. Microculture on cor n-meal agar supplemented with tween 80 showed the presence of pseudomycelium, blastospores and chlamydospores in Candida albicans. The presence of residue and sediment was observed in Sabouraud dextrose broth. White-yellowish, opaque and globose giant Candida colonies were observed. Topographically, the colonies were smooth, wrinkled and fringed at the center and periphery. Assimilation and fermentation of sugars occurred according to each species of

Candida. Proteinase production was detected in all yeast samples analyzed, but phospholipase was only found in Candida albicans. Nine different morphotypes (0000, 3220, 5220, 5230, 5240, 5320, 5330, 5530 and 7330) and 3 different biotypes (377, 677 and 777) were obtained by phenotypic analysis of the yeasts. Fifty-six strains of Candida albicans were classified as serotype A. All Candida species were susceptible to amphotericin B and nystatin. Candida albicans was sensitive to ketoconazole (35.0%), fluconazole (22.8%) and itraconazole (47.4%).Candida krusei and the

Candida tropicalis, however, were not sensitive to fluconazole or cetoconazole, respectively. The Kendall rank correlation test was used to determine the correlation between exoenzyme production by Candidastrains and their in vitro

susceptibility to antifungal drugs.
