4 Material e método

6.3 Informaçoes adicionais

Apesar deste não ser um dos objetivos deste estudo, durante a execução dos experimentos com as soluções MTAD® e Tetraclean®, observamos a tendência destas substâncias alterarem sua

coloração quando armazenadas por períodos de tempo prolongados.

A partir desta observação, foi realizado um teste adicional, onde preparamos as soluções MTAD®, MTAC e Tetraclean®, e armazenamos os tubos em locais expostos à luz, ou protegidos de qualquer fonte de luz. A cada dia foi possível observar o escurecimento gradual das soluções, independente do local onde estavam armazenadas, sendo que o escurecimento foi levemente mais rápido nos tubos expostos à claridade. Após 2 semanas de preparação, as soluções apresentavam coloração em um tom castanho escuro.

Em um segundo momento, observamos que as soluções quando armazenadas em geladeira, a 4oC, não sofriam alteração de cor,

permanecendo límpidas mesmo após 1 mês de preparo. As mesmas soluções foram então armazenadas a 37oC e onde se observou o rápido

escurecimento das soluções. Após 48 horas as soluções já apresentavam um tom castanho escuro semelhante aos das soluções expostas à luz por duas semanas. Imagens das soluções com alteração em sua coloração estão expostas nos Anexos.

Clinicamente, a alteração de cor sofrida pelas soluções poderia determinar a pigmentação da dentina, uma vez que a doxiciclina adere às paredes dentinárias e é eliminada gradualmente7. Também Tay

et al.152 (2007) relataram a ocorrência de uma coloração arroxeada no tecido dentinário de canais radiculares irrigados com NaOCl seguido de MTAD®.

Os resultados obtidos neste estudo reforçam a dúvida quanto à validade do uso de soluções contendo antibióticos em sua formulação. Nossos achados deixaram claro que a ação antibacteriana das soluções Tetraclean® e MTAC decorre principalmente do detergente

cetramida, e não da doxiciclina.

Somado a isto, a alteração cromática sofrida pelas soluções contendo doxiciclina pode determinar uma contra-indicação ao emprego clinico do MTAD® e do Tetraclean®, justificando a continuidade de

pesquisas para o desenvolvimento de soluções irrigadoras com propriedades físico-químicas e biológicas satisfatórias.

7 Conclusão

De acordo com a metodologia empregada neste estudo podemos concluir que:

1. a solução Tetraclean® apresentou melhor ação antibacteriana in

vitro sobre a cepa de E. faecalis VP3-181 e sobre o biofilme que o MTAD®;

2. as variações da fórmula do MTAD®, contendo cetramida em sua composição apresentaram melhor ação antibacteriana in vitro sobre a cepa de E. faecalis VP3-181 e sobre o biofilme que o MTAD®em sua fórmula original;

3. as formulações contendo cetramida e sem conter Tween 80 em sua composição apresentaram melhor ação antibacteriana in vitro sobre a cepa de E. faecalis VP3-181 e sobre o biofilme.
