1. Isolation of Plasmids from E. coli by Alkaline Lysis
Purification of plasmid DNA from Escherichia coli using alkaline lysis (1,2) is
based on the differential denaturation of chromosomal and plasmid DNA in order to separate the two. Bacteria are lysed with a solution containing sodium dodecyl sulfate (SDS) and sodium hydroxide. During this step, chromosomal as well as plasmid DNA are denatured. Subsequent neutralization with potassium acetate allows only the
covalently closed plasmid DNA to reanneal and to stay solubilized. Most of the chromosomal DNA and proteins precipitate in a complex formed with potassium and SDS,
which is removed by centrifugation. The plasmid DNA is concentrated from the supernatant by ethanol precipitation. Using this procedure, 2–5 μg of DNA can be obtained from a 1.5-mL culture of E. coli containing a pBR322-derived plasmid, and three- to five-fold higher yields can be expected from pUC-derived plasmids (3).
2. Isolation of Plasmids from E. coli by Boiling Lysis
The boiling lysis procedure (1) is quick to perform and, therefore, especially suitable for screening large numbers of small-volume Escherichia coli cultures. It is
described with different adaptations in a variety of protocol books (2,3). The quality of the isolated plasmid DNA is lower than that from an alkaline lysis miniprep, but it is sufficient for restriction analysis.
The bacteria are lysed by treatment with lysozyme, Triton, and heat. The chromosomal DNA remains attached to the bacterial membrane and is removed by centrifugation. The plasmid DNA remains in the supernatant from which it is then precipitated
3. Isolation of Plasmids from E. coli by alkaline lysis followed by anion-exchange resin
Plasmid purification protocols are based on a modified alkaline lysis procedure,
followed by binding of plasmid DNA to a anion-exchange resin under appropriate low-salt and pH conditions. RNA, proteins, dyes, and low-molecular–weight impurities are removed by a medium-salt wash. Plasmid DNA is eluted in a high-salt buffer and then concentrated

• Perto da chama transferir cerca de 1,5 mL de cultura bacteriana para um tubo de 1,5
mL estéril
• Centrifugar à velocidade máxima durante 1 minuto para recolher as bactérias no
fundo do tubo
• Ressuspender o sedimento em 200 μL de STET e adicionar 4 μL de lisozima (50
mg/mL)
• Incubar à temperatura ambiente durante 5 minutos
• Ferver durante 45 segundos
• Centrifugar durante 5 minutos à velocidade máxima
• Remover o sedimento com um palito ou ponta estéril
• Adicionar 200 μL de isopropanol e agitar o tubo
• Centrifugar à velocidade máxima durante 10 minutos
• Eliminar o sobrenadante e deixar secar até desaparecer o odor do isopropanol,
evitando que o sedimento seque em demasia
• Ressuspender em 30 – 40 μL de água destilada estéril agitando a base do tubo com
os dedos
