O conhecimento do potencial reprodutivo de um macho é de importância econômica e reprodutiva, principalmente se ele é utilizado em programa de inseminação artificial. Os testes convencionais que avaliam a qualidade seminal não possuem a capacidade de medir o potencial fertilizante de uma amostra e, apenas indicam se a mesma se encontra dentro dos parâmetros aceitáveis para a inseminação segundo o Colégio Brasileiro de Reprodução Animal. O teste de penetração espermática in vitro, aparece neste contexto como um teste laboratorial alternativo para categorizar os machos férteis, quanto a sua capacidade de fecundação, pois mimetizam in vitro o que acontece in vivo. No entanto, este teste tem seu uso limitado por ser de difícil execução e por utilizar equipamentos de alto custo. O presente trabalho objetivou encontrar um novo modelo biológico para estudos sobre aspectos reprodutivos de machos e avaliar alternativas para simplificar e diminuir custos para execução do teste: utilização do teste de penetração na membrana perivitelina interna do ovo da galinha (IPVL) e sua associação com parâmetros in vitro e in vivo; utiilização do BTS como meio de incubação do teste de penetração in vitro e utilização de diferentes maneiras de criopreservação de gametas femininos. Foi concluído que: 1) Calomys laucha pode ser utilizado como modelo biológico; 2) é possível utilizar a membrana perivitelina interna como substrato de teste para predizer a fertilidade de machos Calomys laucha; 3) a IPVL possui receptores que permitiram a ligação do espermatozóide suíno, apresentando potencial para ser utilizada em diagnóstico de fertilidade de machos; 4) é viável utilizar BTS como meio para realização do teste de penetração in vitro em ovócito ou IPVL, em banho-maria sem a presença de CO2 e
5) o congelamento de ovários a -20ºC ou o acondicionamento a 5º C dos ovócitos em PBS para realizar o teste de penetração ovocitária in vitro, utilizando o meio mTBM, é uma alternativa para predizer a fertilidade de sêmen suíno.

The knowledge of the reproductive potential of a male has economical and reproductive importance, especially if he is used in artificial insemination programs. The conventional assays for evaluating seminal quality do not have the capacity of measuring the fertilizing potential of a sample, they only indicate if it is within the acceptable parameters for insemination, according to the Brazilian College of Animal Reproduction. Into this context, the in vitro sperm penetration assay presents itself as an alternative laboratorial test to categorize fertile males regarding their fecundation capacity, since they mimic in vitro what happens in vivo. However, this assay has its use limited by the difficulty of execution and by the utilization of high cost equipment. The present work aimed to find a new biological model to study male reproductive aspects and to evaluate alternatives to simplify and to decrease costs of the assay execution: use of the chicken egg inner perivitelline layer (IPVL) penetration assay and its association with in vitro and in vivo parameters; use of BTS as incubation media for the in vitro penetration assay, and female gametes cryopreservation using different methods. It was concluded that: 1) Calomys laucha can be used as a biological model; 2) it is possible to use the inner perivitelline layer as a substrate in assays to predict fertility of Calomys laucha males; 3) the IPVL has receptors that allow swine sperm binding, therefore presents potential for use in male fertility diagnostic tests; 4) it is possible to use BTS as incubation media for the in vitro penetration assay, in oocytes or in IPVL, and in water-bath without CO2, and 5) the
cryopreservation of ovaries at -20ºC or the refrigeration of oocytes in PBS at 5ºC before the execution of the in vitro oocyte penetration assay, using mTBM media, is an alternative to the use of fresh oocytes in tests to predict swine semen fertility. 1. Introdução Geral
Vários estudos foram realizados na busca de métodos capazes de estimar a fertilidade de reprodutores a partir da sua qualidade seminal. No entanto, as avaliações de qualidade seminal através de métodos convencionais (motilidade, vigor e morfologia espermática), apresentam limitada capacidade de identificar diferenças de menor magnitude, entre machos com potencial de fertilidade mais homogênea (GADEA, 2005). Uma vez que tais métodos apresentam baixa associação com a fertilidade e também são bastante influenciados pelas características individuais dos machos doadores de sêmen (XU et al., 1998; POPWELL & FLOWERS, 2004; MACEDO et al., 2006; 2010), é possível que, eventualmente, machos subférteis sejam usados na rotina de programas de inseminação artificial (IA). Desta forma, a busca por testes de diagnóstico in vitro que identifiquem de forma eficiente os reprodutores com maior potencial fertilizante vem se intensificando.
O teste de penetração in vitro (PIV) em ovócitos simula os processos envolvidos no mecanismo de fertilização, já tendo sido usado para sêmen de uma espécie silvestre de roedores (Calomys laucha), utilizando ovócitos de hamster (LASSERRE et al., 2000). O Calomys laucha é um pequeno roedor encontrado na região Sul da América Latina, com relevância em estudos nas áreas de poluição ambiental e virologia, em função de sua importância epidemiológica como reservatório natural do vírus da hantavirose e de protozoários patogênicos a humanos (MILLS et al., 1994; CHILDS et al., 1995). De acordo com dados gerados por nosso grupo de pesquisa e ainda não publicados, esta espécie apresenta um ciclo reprodutivo extremamente rápido, produz ninhadas grandes e apresenta espermatozóides com morfologia semelhante à de outros mamíferos. Tais características poderiam credenciar esta espécie como um modelo biológico relevante para testar biotécnicas reprodutivas com aplicação em espécies de maior porte e de maior interesse econômico. Porém a viabilidade do seu uso como um modelo biológico em estudos sobre fertilidade in vitro ainda não foi estabelecida.
Ainda que tenha sido usado para sêmen suíno, utilizando ovócitos homólogos (MACEDO et al., 2006; 2010), o teste de PIV ainda é relativamente trabalhoso e oneroso, o que inviabiliza o seu uso em centrais de IA, em condições de rotina (BERGER et al, 1996; LARSSON & RODRÍGUES-MARTÍNEZ, 2000; POPWELL & FLOWERS, 2004; MACEDO et al., 2006). O teste de fertilização in vitro, (FIV) também é relevante neste contexto (SELLÉS, et al., 2003; PELÁEZ et al, 2006). Porém, da mesma forma que o teste de PIV, a FIV também se mostra sensível à variação na capacidade de adesão das células espermáticas aos ovócitos, o que reduz sua precisão na observação de diferenças entre machos (BERGER et al, 1996; LARSSON & RODRÍGUES-MARTÍNEZ, 2000). Além disso, a execução do teste de PIV necessita da obtenção de ovócitos frescos, uma vez que, após a estocagem de ovócitos por períodos superiores a 24 h, a capacidade de PIV de espermatozóides suínos é reduzida (MACEDO et al, 2006). As limitações mencionadas acima justificam a avaliação de outros métodos in vitro e também de métodos alternativos para o acondicionamento de gametas femininos. O teste de PIV na membrana perivitelina interna do ovo da galinha já foi usado para sêmen de galos (ROBERTSON & WISHART, 1997), humanos e de outras espécies mamíferas; (BARBATO et al., 1998), porém ainda não testado com sêmen suíno. Assim, seria necessário testar a hipótese de que espermatozóides com maior capacidade de penetração nesta membrana também apresentariam maior capacidade fertilizante, quando em contato com ovócitos homólogos.
O objetivo do presente trabalho foi estabelecer um método para estimar a fertilidade de machos a partir da capacidade de penetração dos espermatozóides de roedores silvestres e suínos na membrana perivitelina interna do ovo da galinha. Os resultados deste teste foram comparados com métodos convencionais de avaliação de qualidade seminal in vitro e com o teste de PIV simplificado em ovócitos de suínos, em ambas as espécies. Os resultados deste teste também foram comparados com fertilidade in vivo, na espécie Calomys laucha, após monta natural. Além de avaliar o uso de ovócitos suínos frescos, resfriados ou extraídos de ovários congelados no teste de PIV.

Este estudo testou o efeito de diferentes concentrações de lactato de cálcio e tempos de 16
incubação sobre a capacidade de adesão e a penetração dos espermatozóides suínos nas 17
membranas perivitelinas (MP) de ovos de galinhas, para estimar a potencial fertilidade dos 18
machos doadores do sêmen. As amostras (12 ejaculados) foram incubadas em TCM com 19
lactato de cálcio a 1,1 e 2,2 µg/mL. Posteriormente, após definida a concentração de 1,1 20
µg/mL de lactato de cálcio, foram comparados três períodos de incubação a 39°C: 10, 15 e 20 21
min. As respostas testadas foram: a taxa de adesão (TA) e o número médio de 22
espermatozóides aderidos (NA) na MP externa e a taxa de penetração (TP) e o número de 23

orifícios (NO) na MP interna. A TA na MP externa foi de 100%, independentemente da 1
concentração e dos períodos testados. Com a concentração de lactato de cálcio de 1,1 µg/mL, 2
NA na MP externa e TP e NO na MP interna foram superiores (P < 0,05). O NA foi superior 3
com incubação por 20 min, em comparação com períodos mais curtos (P < 0,05). Com 4
incubação por 10 min, não houve penetração na MP interna, mas com incubação por 20 min, a 5
TP na MP interna e o NO foram superiores aos obtidos com incubação por 15 min (P < 0,05). 6
A concentração de 1,1 µg/mL de lactato de cálcio e a incubação por 20 min permitem a 7
execução eficiente de testes in vitro usando MP para estimar a potencial fertilidade de machos 8
suínos. 9

This study tested the effect of different calcium lactate concentrations and incubation periods 14
on the binding and penetration capacity of swine sperm in the perivitelline layers (PL) of 15
chicken eggs, to estimate the potential fertility of the boars used as sperm donors. Semen 16
samples (12 ejaculates) were incubated in TCM medium with two calcium lactate 17
concentrations: 1.1 and 2.2 µg/mL. Then, at a fixed calcium lactate concentration (1.1 18
µg/mL), three incubation periods at 39°C were compared: 10, 15 and 20 min. The tested 19
responses were: binding rate (BR) and number of bound sperm (NB) in the outer PL, and 20
penetration rate (PR) and number of holes (NH) in the inner PL. The BR in the outer MP was 21
100%, regardless of the tested concentrations and incubation periods. The NB in the outer PL 22
and the PR and the NH in the inner PL were greater with 1.1 µg/mL calcium lactate than with 23
2.2 µg/mL. The NB in the outer PL was greater with incubation for 20 min than with shorter 24
periods (P < 0.05). With incubation for 10 min, there was no penetration in the inner PL, but, 25
with incubation for 20 min, the PR and the NH in the inner PL were greater than those with 26
incubation for 15 min (P < 0.05). With 1.1 µg/mL calcium lactate and incubation for 20 min, 1
the in vitro test using PL can be efficiently conducted to estimate the potential fertility of 2

As avaliações convencionais de qualidade seminal (motilidade, vigor e morfologia 8
espermática) são relativamente eficientes, quando utilizadas para triagem de machos com 9
potencial fertilidade muito inferior, mas apresentam limitada capacidade de identificar 10
diferenças de menor magnitude, entre machos de potencial fertilidade mais homogênea 11
(GADEA, 2005). Portanto, machos subférteis podem, eventualmente, ser utilizados na rotina 12
de programas de inseminação artificial (IA). Além disso, estas avaliações apresentam baixa 13
associação com a fertilidade e são sensíveis a variações características individuais atribuídas 14
aos machos doadores de sêmen (XU et al., 1998; POPWELL & FLOWERS, 2004; MACEDO 15
et al., 2006). Portanto, existe a necessidade de desenvolvimento de métodos in vitro eficientes 16
no diagnóstico do potencial fertilizante de machos. 17
O potencial fertilizante de suínos machos pode ser estimado pela capacidade de 18
penetração espermática in vitro em ovócitos homólogos (LARSSON & RODRÍGUES-19
MARTÍNEZ, 2000; POPWELL & FLOWERS, 2004; MACEDO et al., 2006; 2010) ou pela 20
capacidade de fertilização in vitro (PELÁEZ et al, 2006). Entretanto, estes testes são 21
relativamente trabalhosos e onerosos, inviabilizando a sua utilização na rotina de centrais de 22
IA, justificando assim o desenvolvimento de outros métodos de avaliação in vitro. 23
Uma alternativa para a seleção de machos com maior potencial fertilizante é o teste 24
de penetração na membrana perivitelina (MP) interna de ovos de galinhas, utilizado para 25
sêmen de galos (ROBERTSON & WISHART, 1997) ou o teste de adesão na MP externa, já 1
utilizado para sêmen de humanos e de camundongos (BARBATO et al., 1998). 2
Para a realização eficiente de testes in vitro, o meio de co-incubação deve conter 3
componentes que promovam a manutenção e a capacitação dos espermatozóides, a reação 4
acrossomal e a fertilização. O lactato de cálcio é capaz de estimular a capacitação espermática 5
e/ou a reação acrossomal em espermatozóides suínos (ABEYDEERA & DAY, 1997). Ainda, 6
há uma grande variação no tempo de co-incubação dos gametas, ainda que a maioria dos 7
protocolos utilize co-incubação por 6 h (ABEYDEERA & DAY, 1997; MACEDO et al., 8
2006). Entretanto, Gil et al. (2004) observaram que, com o períodos de co-incubação entre 10 9
e 30 min, havia um aumento no número de espermatozóides suínos por ovócito penetrado. 10
Porém, uma vez que os testes de adesão e penetração in vitro nas MP de ovos de galinhas 11
ainda não foram conduzidos com sêmen suíno, é necessário determinar qual seria a 12
concentração de lactato de cálcio e o período de incubação mais eficientes para a condução 13
deste teste. 14
O objetivo deste estudo foi testar o efeito de diferentes concentrações de lactato de 15
cálcio e períodos de incubação sobre a capacidade de adesão e penetração in vitro de 16
espermatozóides suínos nas MP interna e externa de ovos de galinhas. 17

MATERIAL E MÉTODOS 19
As amostras de sêmen avaliadas neste experimento (12 ejaculados) foram coletadas 20
de três machos suínos sexualmente maduros e com fertilidade conhecida, que estavam em 21
rotina de coleta de sêmen em uma central comercial de IA, localizada no município de Pelotas 22
– RS, sendo coletados duas vezes por semana durante um mês. Imediatamente após a coleta, 23
os ejaculados foram diluídos em Beltsville Thawing Solution (BTS), com concentração de 3,5 24
x 109 espermatozóides/100 mL (PURSEL & JOHNSON, 1975) e enviados para o laboratório. 25
Somente foram utilizados ejaculados com motilidade igual ou superior a 80%. Os parâmetros 1
de qualidade seminal avaliados logo após a diluição do sêmen foram: motilidade e morfologia 2
espermática. Todos os químicos utilizados neste experimento são de procedência da Sigma 3
Chemical Company (St. Louis, MO – USA). 4
A motilidade espermática foi avaliada através da microscopia ótica, sob aumento de 5
200 x (BEARDEN & FUQUAY, 1997), sempre pelo mesmo técnico treinado. A morfologia 6
espermática foi avaliada segundo o protocolo de Hancock & Hovell (1959), com a contagem 7
de 200 células em microscopia óptica com contraste de fase. 8
As MP eram provenientes de ovos de galinhas, frescos e não férteis, tendo sido 9
isoladas conforme descrito por Robertson & Wishart (1997). No primeiro experimento, foi 10
utilizado meio composto por: TCM 199; 0,91mM de piruvato; 5,5 mM de glucose; 50 µg/ml 11
de sulfato de estreptomicina; e lactato de cálcio em duas concentrações (1,1 ou 2,2 µg/mL). 12
Este meio foi descrito anteriormente por Macedo et al. (2006), para o teste de penetração in 13
vitro de sêmen suíno em ovócitos homólogos, Aproximadamente 10 min antes do teste, os 14
ejaculados foram submetidos a centrifugação a 900g por 3 min, para remover o BTS e não 15
alterar o meio de fertilização. A concentração espermática do pellet foi determinada e o 16
volume de sêmen correspondente à 1 x 106 espermatozóides por mL foi adicionado 17
diretamente em 750 µL do meio de fertilização. As MP foram incubadas com os 18
espermatozóides em banho-maria a 39°C, sem atmosfera controlada e sem agitação. No 19
primeiro experimento, a incubação durou 20 min. 20
No segundo experimento, foi utilizado lactato de cálcio na concentração de 1,1 21
µg/mL, conforme indicado pelos resultados do primeiro experimento. Foram comparados três 22
períodos de incubação: 10, 15 e 20 min. Após estes períodos, as MP foram lavadas para a 23
retirada dos espermatozóides não aderidos e esticadas sobre uma lâmina. O corante 24
fluorescente Hoechst 33342 foi adicionado sobre a MP externa, para a leitura da lâmina. A 25
contagem do número de espermatozóides aderidos à MP externa foi realizada sob luz 1
ultravioleta (filtro de excitação BP 330 - 385), em microscópio de epifluorescência, sob 2
aumento de 200 X. 3
As lâminas contendo as MP internas foram observadas em microscópio de campo 4
escuro, sob aumento de 200 X. Três campos do microscópio foram fotografados, para a 5
contagem do número de pontos de orifícios, representado os pontos de hidrólise na MP 6
interna. Posteriormente, foi calculado o número médio de orifícios por mm2. 7
Estatísticas descritivas foram calculadas para motilidade e morfologia espermática. 8
Comparações entre os números de espermatozóides aderidos à MP externa e penetrando a MP 9
interna em função das concentrações de lactato e dos tempos de incubação foram comparados 10
através da análise de variância Kruskal-Wallis. Comparações das taxas de adesão na MP 11
externa e de penetração na MP interna foram feitas pelo teste de qui-quadrado. As análises 12
estatísticas foram realizadas através do software Statistix (2003). 13

As médias para motilidade e morfologia espermática normal observadas nas amostras 17
de sêmen coletadas para este experimento foram de 90,0 ± 5,3 e 94 ±2,3, respectivamente. O 18
número médio de espermatozóides aderidos na membrana perivitelina externa foi de 37,5 ± 19
35,8 e o número de orifícios na membrana perivitelina interna por mm2 foi igual a 5,2 ± 2,9 20
A concentração de 1,1 µg/mL de lactato de cálcio apresentou respostas superiores às 21
observadas com a concentração de 2,2 µg/mL, tanto para a MP externa, quanto para a MP 22
interna (Tabela 1). No presente estudo, a maior concentração de lactato de cálcio influenciou 23
negativamente o resultado dos testes com ambas as MP, o que pode ser atribuído à maior 24
concentração de cálcio, que pode induzir precocemente a capacitação espermática e a reação 25
acrossomal, antes da interação com as proteínas da zona pelúcida (ABEYDEERA & DAY, 1
1997). 2
O número de espermatozóides aderidos na MP externa e o número de orifícios na MP 3
interna foram maiores com incubação por 20 min (Tabela 2). A avaliação da capacidade de 4
adesão espermática na MP externa dos ovos de galinha não se mostrou útil para discriminar 5
diferenças em fertilidade entre indivíduos, pois apresentou resultados positivos em 100% das 6
amostras, independentemente da concentração de lactato de cálcio (Tabela 1) e do período de 7
incubação (Tabela 2). Estes dados sugerem que os espermatozóides não necessitam sofrer 8
reação acrossomal para se aderirem na MP externa, o que está em concordância com os 9
achados de Fazeli et al. (1997), que relatam que 8% dos espermatozóides aderidos a ovócitos 10
apresentavam o acrossoma íntegro, mesmo após co-incubação por 6 h. 11
O teste de penetração espermática in vitro na MP interna pode ser considerado mais 12
eficiente do que o teste com a MP externa, por refletir não somente a capacidade de adesão do 13
espermatozóide, mas também sua habilidade em sofrer a reação acrossomal e hidrolisar a 14
membrana. Os resultados obtidos com o tempo de incubação por 20 min podem viabilizar a 15
adequação do teste de penetração in vitro na MP para ser usado na rotina em centrais de IA, 16
em função de sua rápida execução e por necessitar a utilização de equipamentos que estão 17
normalmente disponíveis nestas instalações. Por outro lado, sua utilização nestas condições 18
deve ser economicamente viável e deverá ser validada, em estudos futuros, através de 19
comparações entre os resultados observados in vitro com a fertilidade in vivo. 20 21 CONCLUSÕES 22 23 21

interna de ovos de galinhas pode ser realizado de forma eficiente com o uso do meio TCM 2

As Calomys laucha rodents present a short reproductive cycle and sperm morphology 12
similar to domestic species, they may be used as a biological model for in vitro fertilization 13
studies. This study evaluated sperm quality of Calomys laucha by conventional methods, 14
tested improvements in the in vitro penetration (IVP) test of Calomys laucha sperm in 15
swine oocytes and in perivitelline layers (PVL) of chicken eggs, and evaluated the 16
associations and the accuracy of the IVP in comparison with in vivo fertility after natural 17
mating. Evaluations of sperm quality were compared as a function of the IVP in swine 18
oocytes and in PVL and to in vivo fertility. The sensitivity and specificity of both IVP tests 19
were determined considering in vivo fertility as the gold standard. Correlations were 20
measured by Spearman rank correlation. Calomys laucha sperm presented motility, normal 21
morphology, membrane integrity and acrosome integrity equal to 90.6 ± 5.6%, 90.2 ± 22

6.6%, 88.7 ± 9.6% and 90.5 ± 11.5%, respectively. The IVP rate was 39.8% in swine 23
oocytes and 87.5% in the inner PVL. The IVP in swine oocytes presented sensitivity of 24
16.0% and specificity of 83.3%. The sensitivity of the IVP in the inner PVL was 84.0%, but 25
the specificity was not determined because there were no true negative results. Sperm 26
membrane integrity was positively correlated with parturition after natural mating (r = 0.38, 27
P < 0.01) and litter size (r = 0.54; P < 0.0002). Calomys laucha may be used as a biological 28
model for in vitro fertilization studies. 29

Conventional methods of semen quality evaluation, such as sperm motility, 32
morphology and concentration, are routinely used to estimate male reproductive potential in 33
domestic animal species, even though their correlations with actual fertility are 34
unsatisfactory (Barth, 1992). To overcome such limitations, the capacity of spermatozoa to 35
penetrate homologous and heterologous oocytes in vitro can be used to estimate fertility 36
(Zhao et al., 2002; Macedo et al., 2006; 2010). In boars, the in vitro penetration (IVP) test 37
in homologous oocytes detected a decline in sperm penetrating capacity after only 24 h of 38
storage, which was undetected by conventional semen quality evaluations after storage for 39
72 h (Macedo et al., 2006). However, the IVP test was considered expensive and labor 40
intensive because it required freshly recovered oocytes and a CO2 incubator for gametes
co-41
incubation. Advances in the IVP test were achieved by using oocytes frozen in cryotubes 42
and also flask or bag systems for gametes co-incubation without a CO2 incubator (Macedo
43
et al., 2010), but such test still needs further improvement to be feasible for routine use. 44
Furthermore, the IVP test is highly sensitive to individual variation in the sperm penetrating 45
capacity in oocytes among the males used as sperm donors (Berger et al., 1996; Larsson 46
and Rodríguez-Martínez, 2000; Popwell and Flowers, 2004; Macedo et al., 2006; 2010). 47
Considering the orthologous similarity among glycoproteins from the zona pellucida 48
of mammal’s oocytes (mice, ovine, swine and human) and proteins from the inner 49
perivitelline layer (PVL) of chicken eggs (Takeuchi et al., 1999; Wassaman, 1999; Smith et 50
al., 2005; Mugnier et al., 2009), male’s potential fertility can be estimated by the sperm IVP 51
in the inner PVL, assuming that it would be related to sperm IVP in oocytes. The IVP in the 52
inner PVL was used to estimate potential fertility of rooster sperm (Robertson and Wishart, 53
1997), whereas the IVP in the whole PVL was used for sperm of mice, horses, rams, bulls 54
and humans (Barbato et al., 1998). 55
Calomys laucha are small rodents found in South America inhabiting dunes in
56
which they are exposed to extreme temperature variation (0° to 50° C) throughout the year 57
(Camargo et al., 2006). Such rodents have epidemiologic relevance as natural reservoirs of 58
the Hantavirus and of protozoa that are pathogenic to humans (Mills et al., 1994; Childs et 59
al., 1995). For such species, the estrous cycle lasts 5 to 6 d, gestation lasts 21 d, litters have 60
up to 8 products (Mills et al., 1992) and sperm morphology is similar to that from other 61
mammals (Lassere et al., 2000). Thus, such characteristics may allow Calomys laucha to be 62
a relevant biological model for in vitro fertilization studies aimed to develop techniques to 63
be used in species of economic interest. However, the usefulness of such species as a 64
biological research model is still unknown due to the lack of evaluations of semen quality 65
and potential in vitro fertility. The IVP test for Calomys laucha sperm using hamster 66
oocytes incubated in TALP medium including 4% BSA presented great variation due to the 67
sperm concentration used for oocyte’s insemination and to the time of sperm capacitation 68
(Lassere et al., 2000). As the zona pellucida of swine oocytes can be penetrated by sperm 69
from bulls and rats (Zhao et al., 2002), it is possible to evaluate the potential fertility of 70
Calomys laucha sperm through the IVP test using swine oocytes, which was not done yet.

Also, the IVP test in PVL was not yet conducted for Calomys laucha sperm. 72
The objectives of this study were: to describe sperm quality of Calomys laucha after 73
evaluation by conventional methods; to test improvements in the IVP test in both swine 74
oocytes and in PVL using Calomys laucha males as sperm donors; and to evaluate the 75
associations among sperm quality and potential fertility of Calomys laucha with in vivo 76
fertility after natural mating with Calomys laucha females, as well as the accuracy of the 77
IVP tests in comparison with in vivo fertility. 78

Materials and methods

Sperm collection and evaluation

The Calomys laucha males and females evaluated in this study were from the 82
vivarium of non-conventional animals of the Universidade Federal de Rio Grande, RS, 83
Brazil. The animals were housed in plastic boxes (35 x 20 x 13 cm), kept in an environment 84
with controlled temperature (20.0 ± 2.0 ºC) and photoperiod (LD 12:12, with light on at 85
6:00 a.m. and off at 6:00 p.m.), and received food and water ad libitum. The procedures 86
used for experimentation and euthanasia of the animals followed the recommendations of 87
the Brazilian law # 6638 of May 8, 1979, and also those of the U.S. National Institute of 88
Health guide for the use of laboratory animals (1996) and of Gannon et al. (2007). 89
Sperm donors were 32 three-month old Calomys laucha males. As the testes were 90
internal, sperm was collected after approaching the reproductive system by laparotomy. 91
Then, the tail of both epididymis and part of the vas deferens were removed, ruptured with 92
a 12 g x 40 mm needle and placed in a 35 mm Petri dish containing 750 µL of M2 medium 93
with HEPES (M7167) (Maleszewski et al., 1995), for sperm dilution. All chemicals used in 94
this experiment were from Sigma Chemical Company (St. Louis, MO, USA). 95
Sperm quality evaluations were done after incubation of samples for 10 min at 37ºC 96
in M2 medium. Sperm motility was evaluated by optical microscopy at 200 x (Sztein et al., 97
2000), always by the same trained technician. Sperm morphology was determined as 98
described by Tayama et al. (2006), by counting 200 cells with phase contrast microscopy at 99
200 x. A Neubauer chamber was used to determine the sperm concentration to be used in 100
the IVP tests. The evaluations of sperm membrane and acrosome integrity and the IVP in 101
oocytes and in the PVL were conducted with an epifluorescent microscope (Olympus BX 102
51, América INC, São Paulo - Brasil), with filter wave length of 450-520 nm. Sperm 103
membrane integrity was evaluated using carboxyfluorescein diacetate (CFDA; C5041) and 104
propidium iodide (PI; P4170) (Harrison and Vickers, 1990), at 400 x. In each slide, 200 105
cells were counted and classified as intact (green fluorescence) or not intact (red 106
fluorescence or simultaneous red and green fluorescence). Acrosome integrity was 107
evaluated using FITC-PNA (L7381) by counting 200 cells in dry slides. Acrosomes were 108
classified as intact, when presented red fluorescence and normal conformation, or not 109
intact, when presented green fluorescence or no fluorescence and conformation distinct 110
from that from normal spermatozoa (Jiménez et al., 2003). 111

IVP in immature swine oocytes

Oocytes were collected from prepubertal gilts from an abattoir and transported to 114
the laboratory in saline + gentamicin (40 mg/mL) at 30 ◦C within 60 min. Upon arrival, 3–6 115
mm follicles were aspirated using a 12 g x 40 mm needle attached to a 10 mL syringe. 116
Further oocyte’s processing followed procedures described by Macedo et al. (2006; 2010). 117
For each Calomys laucha male, 30 swine oocytes were used. The test was conducted with 118
the protocol described by Maleszewski et al. (1995) with modifications, using M2 with 119
HEPES containing 0.4% BSA (A3311) as the fertilization medium. Gametes co-incubation 120
was done in water bath at 37ºC for 2 h (Lassere et al., 2000), using a concentration of 2 x 121
106 spermatozoa/mL. After co-incubation, oocytes were recovered, washed, stained with 122
Hoescht 33342 (10 µg/mL) and evaluated in an epifluorescent microscope at 400 x. The 123
number of penetrated oocytes (having zona pellucida containing at least one spermatozoon, 124
some of which had elongated and swollen heads) and of spermatozoa per oocyte were 125
recorded (Lassere et al., 2000; Macedo et al., 2006; 2010). For each male, the IVP rate was 126
calculated as described by Macedo et al. (2006; 2010). Subsequently, each male was 127
classified according to its mean IVP rate (0: on average or at most one standard deviation 128
below average; 1; at least one standard deviation above average). 129

Both the outer and the inner PVL were obtained from fresh and non fertile chicken 132
eggs, which were isolated as described by Robertson and Wishart (1997). The medium and 133
co-incubation protocol were the same described above for the IVP test in swine oocytes. 134
The tests were conducted in duplicate, using two layers per sperm sample (n = 64, for both 135
outer and inner PVL). After co-incubation, the PVL were washed for removal of loosely 136
attached spermatozoa and stretched over a slide. The outer PVL was stained with Hoechst 137
33342 and the spermatozoa bound to it were counted, using an epifluorescent microscope at 138
200 x. The inner PVL was evaluated using a dark field microscope at 200 x. After 139
photographing three fields of each slide, the number of holes (representing the hydrolysis 140
points made by sperm penetration in the PVL) was counted. Then, the mean number of 141
holes per mm was calculated. Calomys laucha males were classified according to the IVP 142
in the inner PVL (0: layer having no penetration; 1: at least one penetrated layer). 143

In vivo fertility

Prior to sperm collection, each Calomys laucha male was mated with one adult 146
female. Each pair of animals should have known fertility: they were mated previously to 147
the experiment and produced at least one litter. Each pair was kept together for 7 d and the 148
mating was recorded when a vaginal plug was observed in the females. After 21 d, the 149
occurrence of parturition and the litter size were recorded. 150

Statistical analyses

Descriptive statistics were calculated for: sperm motility, morphology, membrane 153
and acrosome integrity; number of sperm bound to the outer PVL, of holes in the inner 154
PVL and of sperm penetrating in swine oocytes; and litter size after natural mating. Those 155
variables were compared as a function of occurrence of parturition after natural mating and 156
to the IVP in both swine oocytes and in the inner PVL by analysis of variance of Kruskal-157
Wallis for non parametric data. Comparisons among occurrence of parturition after natural 158
mating and IVP in swine oocytes and in the PVL were done by the Fischer’s exact test. The 159
sensitivity and the specificity of the IVP in swine oocytes and in the inner PVL were 160
calculated to determined their accuracy (Dohoo et al., 2003), considering the occurrence of 161
parturition after natural mating as the gold standard. Sensitivity and specificity were also 162
calculated for the IVP in the inner PVL considering the IVP in swine oocytes as the 163
standard. Correlations among the variables mentioned above were determined by Spearman 164
rank correlation coefficients. All analyses were conducted with Statistix® (2008).

Results

Mean motility, normal morphology and membrane integrity for Calomys laucha 168
sperm were equal to 90.6 ± 5.6%, 90.2 ± 6.6% and 88.7 ± 9.6%, respectively. Mean 169
acrosome integrity was 90.5 ± 11.5%. 170
Considering the 960 swine oocytes used, the IVP rate was 39.8%. Penetration by 171
multiple sperm occurred in 58.2% of the penetrated oocytes. Only 5 of the 32 tested males 172
presented above-average IVP rate.The only parameter that differed for males with distinct 173
IVP in swine oocytes was acrosome integrity (Table 1), which was greater for those with no 174
IVP (P < 0.05). The number of penetrating spermatozoa per oocyte was 2.5 ± 2.2. 175
As the sperm binding rate in the outer PVL was 100%, such layer was not included 176
in subsequent in vitro tests. The number of spermatozoa bound to the outer PVL was 531.4 177
± 466.8. In the inner PVL, the IVP rate was 87.5% and the number of holes by mm2 was 178
1.9 ± 1.8. Only 4 males did not present IVP in the inner PVL. None of the evaluated 179
parameters differed (P > 0.05) for males with distinct IVP in the inner PVL (Table 1). 180
After natural mating, the parturition rate of Calomys laucha females was 81.3% and 181
litter size was 4.3 ± 1.3. In comparison with the males that did not sire a litter after natural 182
mating, those that did sire a litter presented greater sperm membrane integrity (P < 0.05), 183
but lower normal sperm morphology (P < 0.05) (Table 1). 184
Considering in vivo fertility as the gold standard, the IVP in swine oocytes presented 185
sensitivity and specificity of 16.0% and 83.3%, respectively (Table 2). The sensitivity of 186
IVP in the inner PVL was 84.0%, but it was not possible to calculate the specificity because 187
none of the tested males presented negative results for both tests simultaneously. When the 188
IVP in swine oocytes was considered as the standard, the IVP in the inner PVL presented 189
sensitivity of 80.0% and specificity of 11.1% (Table 2). 190
Correlations among parameters of sperm quality, IVP in swine oocytes and in PVL 191
and in vivo fertility are shown in Table 3. Sperm membrane integrity was positively 192
correlated with both the occurrence of parturition after natural mating and litter size (P < 193
0.05). Sperm motility did not correlate with any of the evaluated parameters (P > 0.05). 194

Discussion

This was the first study to conduct a detailed reproductive evaluation of Calomys 197
laucha males, considering sperm quality and IVP in PVL and in heterologous oocytes from
198
non rodent species. The parturition rate observed in this study can be considered acceptable 199
for animals maintained in a controlled environment. However, during their reproductive 200
season (in the summer) in their natural environment, Calomys laucha may achieve 201
parturition rates of 100%, even when exposed to great temperature variation inside the 202
dunes where they inhabit (Mills et al., 1992). The present study generated knowledge about 203
the reproductive physiology of such species, which may be helpful for their preservation. 204
Those results also suggest that Calomys laucha may be a useful biological model for in 205
vitro fertilization studies intended to produce knowledge for species of economic interest.

As the sensitivity of the IVP in the inner PVL was 84.0%, 16.0% of the tested males 207
were false negatives, since they produced a litter after natural mating even though they 208
failed to penetrate the inner PVL in vitro. On the other hand, 84.0% of the tested males 209
were false negatives according to the IVP test using swine oocytes. Thus, considering in 210
vivo fertility as the gold standard for both tests, the IVP test was more sensitive using the
211
inner PVL than using swine oocytes. Although both IVP tests lacked sensitivity, it is 212
important to emphasize that such tests are used to estimate male’s potential fertility 213
together with a greater group of other evaluations (Van der Ven et al., 1986). 214
As the IVP in swine oocytes presented high specificity, such test was accurate on 215
detecting sub fertile males unable to sire a litter after natural mating by detecting those 216
failing to penetrate swine oocytes in vitro. However, as no male failed on both penetrating 217
the inner PVL and siring a litter, it was impossible to determine the test’s specificity. The 218
evaluation of the sperm binding to the outer PVL was not useful to determine potential 219
male fertility because sperm from all males bound to the outer PVL, regardless of their 220
performance in the other tests. The low specificity of the IVP test in the inner PVL when 221
the IVP on swine oocytes was considered as the standard suggests that the accuracy on 222
detecting lowly fertile males may actually be limited for both such tests. Ideally, in vitro 223
tests to identify sub fertile males should present low false positive rate in a small sample 224
size, considering the cost-benefit ratio and the time needed to evaluate all parameters of 225
sperm quality in such tests (Van der Ven et al., 1986; Barbato et al., 1998). The low 226
specificity of the IVP test in the inner PVL when in vivo fertility was the gold standard may 227
not necessarily reflect male sub fertility, but instead reproductive disorders that prevented 228
females to conceive or to maintain pregnancy, since all the six males that failed to sire a 229
litter after natural mating presented IVP in the inner PVL.

Some studies reported moderate correlation between IVP in oocytes and in vivo 231
fertility (Hammitt et al., 1989; Berger and Parker, 1989; 1996; Gadea et al., 1998). 232
However, in this study, the IVP in both swine oocytes and in the inner PVL were not 233
correlated with each other and also uncorrelated with in vivo fertility, which agrees with 234
other studies (Gadea et al., 1998; Tardif et al., 1999; Braundmeier et al., 2002; Collins et 235
al., 2008), but contradicts reports of correlation between in vivo fertility and IVP of human 236
sperm in hamster oocytes (Van der Ven et al., 1986). Such lack of correlation may have 237
been due to gametes’ co-incubation in M2 medium without 5% CO2, which may have
238
failed to provide an adequate environment for in vitro sperm capacitation. That may also be 239
related with the negative correlation between the IVP in swine oocytes and acrosome 240
integrity. Acrosome integrity was also negatively correlated with the number of penetrating 241
spermatozoa per oocyte and presented no correlation with parturition after natural mating, 242
which partially agrees with the low correlations observed among such variables in other 243
studies (Hammitt et al., 1989; Gadea et al., 1998; Soler and Garde, 2003). 244
Normal sperm morphology was greater than 85.0%, for all tested Calomys laucha 245
males. Thus, even those that failed to sire a litter after natural mating would be considered 246
fertile, according to breeding soundness evaluation criteria used for most mammalian 247
species. Nonetheless, although normal sperm morphology was negatively correlated with in 248
vivo fertility, it should be emphasized that normal sperm morphology does not necessarily
249
indicate normal sperm functionality (Bonet and Briz, 1991), which is supported by the fact 250
that Calomys laucha males siring a litter after natural mating also presented higher sperm 251
membrane integrity than those that did not sire a litter, as reported by Gadea et al (1998) for 252
swine. Therefore, sperm membrane integrity may be a relevant parameter to estimate 253
potential fertility of Calomys laucha males. On the other hand, sperm membrane integrity 254
was negatively correlated with the IVP in swine oocytes in the present study and presented 255
no correlation with IVP in oocytes in other species, when determined by the hyposmotic 256
swelling test (Tardif et al., 1999; Van der Ven et al., 1986; Soler and Garde, 2003). 257
The IVP rate in swine oocytes observed in the present study (39.8%) indicate that 258
oocytes from slaughtered females can be used as substrates for in vitro fertilization studies, 259
allowing a reduction in the need of euthanasia of laboratory animals commonly used as 260
oocyte’s donors. Furthermore, the high IVP rate previously reported for Calomys laucha 261
sperm in hamster’s oocytes (Lassere et al., 2000) was obtained with incubation in TALP 262
medium with 4% BSA, after sperm pre-incubation for 90 min (which likely induced sperm 263
capacitation) and gametes co-incubation in 5% CO2 for additional 4 h, thus requiring 5 h
264
and 30 min to be completed. Thus, even though the IVP rate in swine oocytes observed in 265
the present study were apparently lower than those reported elsewhere for swine sperm in 266
homologous oocytes (Gadea et al., 1998; Popwell and Flowers, 2004; Macedo et al., 2006; 267
2010), the present study contributed for improvement in the IVP test in swine oocytes, 268
allowing its execution in nearly 2 h without the need of a CO2 incubator, which may permit
269
the IVP test to be used in commercial artificial insemination studs in the future. 270

Conclusions

In vivo fertility of Calomys laucha males was positively correlated with their sperm

membrane integrity, but uncorrelated with the IVP in both swine oocytes and in the inner 274
PVL of chicken eggs. The IVP test in swine oocytes can be conducted in nearly 2 h without 275
incubation in CO2. Calomys laucha may be a feasible biological model for in vitro
