Resumo
A anemia falciforme é uma doença hereditária com alta incidência na população, causada por um defeito estrutural na cadeia β-globina, originando a HbS. Essa hemoglobina anômala, sob baixas tensões de oxigênio, forma polímeros intracelulares, provocando a deformação dos eritrócitos que por sua vez bloqueiam a microcirculação, ocasionando i n f ar to d e v ár i o s ó r gão s. Estu d o s cl ín i co s demonstraram que a HbF possui a capacidade de inibir a polimerização da UbS e reduzir a falcização, melhorando o quadro clínico.
A i n d u ção f ar m aco l ó gi ca p o r v ár i o s co m p o st o s q u í m i co s, d en t r e o s q u ai s a hidroxiuréia (HU), 5-azacitidina, butirato e os hormônios esteróides, aumentam a síntese de HbF em pacientes com anemia falciforme. A regulação da expressão da UbF está relacionada à presença de elementos ativos ligados em “ cis” ao co m p l ex o d a β-gl o b i n a, b em co m o d e p r o t eí n as r eg u l at ó r i as d o p r o c esso d e tran scri ção gên i ca d en o mi n ad as f ato res d e transcrição, como as proteínas CATA-1, GATA-2 e N F- EGATA-2, ex p r essas p r i n ci p al m en t e n a linhagem eritróide.

Esse projeto teve por objetivo estudar o efeito da HU e do hormônio progesterona na modulação da expressão gênica pelos fatores de transcrição GATA-1, GATA-2 e NF-E2 e do gene γ globina, em células da linhagem eritróide obtidas através da cultura de duas fases a partir de sangue periférico de doadores de sangue aten d i d o s n o H em o cen tr o d a Un i cam p . A expressão dos genes dos fatores de transcrição foi determinada por RT-PCR semiquantitativo e quantificação por densitometria.

Pharmacological induction using various chemical compoun ds, the best kn own bei n g hydroxyurea (HU), 5-azacytidine, butyrate and steroid hormones, increase the synthesis of HbF in sickle cell patients. The regulation of the expression of HbF is related to the presence of active elements linked “ cis” to the beta-globin as well as to the presence of regulatory proteins of the genomic transcription process (known as transcription factors), such as the 1, GATA-2 and NF-EGATA-2 proteins, expressed principally in the erythroid lineage

GATA-1, GATA-2, NF-E2 an d the gamma globi n gen e i n erythroi d li n eage cells obtai n ed from two-phase culture of peri pheral blood don ors at the Hemocen ter , UNICAMP. The expr essi on of the tr an scr i pti on factor s was deter mi n ed by semi -qu a n t i t a t i ve RT-PCR a n d -qu a n t i f i ed by den si tometry.
Ou r r esu lts demon str ate that i n the cells tr eated wi th HU an d pr ogester on e ther e was a si gn i fi can t i n cr ease, when compar ed wi th the u n tr eated cells ( p<0.05, Wi lcoxon test) , i n the expressi on of the GATA-l tran scri pti on factor an d of the gamma globi n gen e after 96hs of phase II culture un ti l termi n ati on at 216hs. The expressi on of the NF-E2 tran scri pti on factor was n ot affected by HU or progesteron e admi n i strati on duri n g the en ti r e pr ocess of er ythr oi d di ffer en ti ati on an d n ei ther of the chem i cals had an y si gn i fi can t i n flu en ce u pon the expr essi on of the GATA-2 tran scri pti on factor (p>O.O5, Wi lcoxon test).
